Its dopamine-releasing action in the nucleus accumbens is weak and
dose-dependent; the likelihood of a euphoric response ('abuse
potential'), dose-escalation and tolerance is thus apparently small.
Modafinil has central alpha 1-adrenergic agonist effects i.e. it
directly stimulates the receptors. Modafinil inhibits the reuptake of
noradrenaline by the noradrenergic terminals on sleep-promoting neurons
of ventrolateral preoptic nucleus (VLPO). More significant, perhaps, is
its ability to increase excitatory glutamatergic transmission. This
reduces local GABAergic transmission, thereby diminishing GABA(A)
receptor signalling on the mesolimbic dopamine terminals.
Cephalon, Takeda To Co-Promote Provigil In US

Cephalon Inc. (CEPH) and Takeda Pharmaceutical North America Inc. will
co- promote Provigil in the U.S.
In July, 500 Takeda sales representatives will begin promoting Provigil,
which is indicated for improving wakefulness. At that time, the Cephalon
and Takeda sales force promoting Provigil will comprise 900 people.
The co-promotion agreement will run for three years with an option to
renew annually. Cephalon, of Frazer, Pa., will retain all responsibility
for the development of Provigil, and pay Takeda a royalty.
Takeda Pharmaceuticals North America, of Liconlnshire, Ill., is a unit
of Takeda Pharmaceutical Co. (4502.TO) of Japan.
